I recently started building this particular Spitfire (EN921, YO-A) using Tamiya's Vb kit and was wondering if somebody can help me with these questions:

Did this particular Vb had the wing strengthening reinforcement strips on top of the wing?
Also, when did these appear on Vb wings?

The overwing stiffeners were introduced, as mod 532, to the production line from July 16, 1942, with leaflets being issued to repair depots, etc., between June 15th, and July 16th.  The work was "Class 2," and could be done outside the company, so, if your model is after that date, it's entirely likely to have had the stiffeners.  Eventually the modded wings were retro-fitted, in fact AR213, a Mk.I, formerly based here, had one wing with, and one without, the stiffeners.

"The bible" lists EN921 as built 14-5-42.  Since the markings I'm going to use are from the Fall of 1943, I think I'll leave them on.  Great news - less work Wink.
